ABSTRACT

Sea buckthorn berries and leaves are two medicinal parts derived from the same primitive plant,mostly used as ethnic medicine,which have a long history in Mongolian and Tibetan. This paper reviews the studies on the chemical composition and differences between sea buckthorn berries and leaves. They have the same or similar composition and content of flavonoids,triterpenes,steroids,organic acids and volatile oils,also have some differences. The main differences are as follows: the flavonoids content in the sea buckthorn leaves is greater than that of the sea buckthorn berries,but the species of flavonoids in the berries are higher than leaves. The species and content of steroids and organic acids in the berries are higher than that of the leaves. The berries contain abundant volatile oil,and the leaves contain many phenolic compounds. Finally,the main problems and the prospect of the next research are put forward.

ABSTRACT

El objetivo de la investigación fue caracterizar y comparar 5 variedades comerciales de arándano y una especie nativa. Para lo cual, las muestras comerciales fueron adquiridas de supermercados de la ciudad de Trujillo y la especie nativa fue recolectada del Distrito de Sonche (Centro Poblado Olmal). Se determinó humedad, acidez, sólidos solubles totales (SST), materia seca, peso, proteína, grasa, cenizas, pH, actividad antioxidante y aceptación con 30 panelistas sin entrenamiento. Las variedades comerciales Biloxi, Misty y Emerald fueron las que alcanzaron los más altos resultados en características fisicoquímicas: humedad, acidez, SÓLIDOS solubles totales, peso, proteína, pH, a diferencia del contenido de: fibra, cenizas, grasas y energía donde la especie nativa sobresalió, superando en valores a las variedades comerciales. Los índices más altos de actividad antioxidante lo alcanzaron las variedades Biloxi, Misty y Emerald. Las variedades con la mejor aceptación fueron Misty y Biloxi alcanzando un puntaje de 8, a diferencia de las otras variedades que obtuvieron un puntaje de 7 al igual que la especie nativa.

Objective To evaluate the variation of sweet chemicals (sugars and betaine) in fruits of six species in genus Lycium L. i.e. Lycium truncatum, L. cylindricum, L. dasystemum, L. dasystemum var. rubricaulium, L. chinense, and L. barbarum harvested at three different ripening stages. Methods A simple and effective method based on UPLC-ELSD was developed for the simultaneous determination of two monosaccharides (glucose and fructose), two disaccharides (sucrose and maltose), one sugar alcohol (xylitol), and betaine in wolfberry fruits (goji berries) of genus Lycium L. Results The six species of wolfberry fruits harvested at the three different ripening stages were evaluated in sugars and betaine contents. Fructose and glucose were the predominant sugars in mature wolfberry fruits. Fructose, glucose, and betaine, as well as total sugar contents, increased continually over the ripening process. L. truncatum and L. dasystemum had higher contents of sugars and betaine than the other species. Conclusion UPLC-ELSD is a simple, reliable and effective method for analysis of the sweet chemicals in wolfberries. Wolfberry fruits at the different ripening stages were significant different in sweetness. L. truncatum and L. dasystemum could be the potential sources of wolfberry fruit sources.

ABSTRACT

O câncer de mama caracteriza-se globalmente como a neoplasia de maior incidência e mortalidade na população feminina. Antocianinas e elagitaninos presentes em frutas como as berries destacam-se por seu promissor efeito protetor em diferentes estágios do desenvolvimento do câncer de mama. Grumixama (G; Eugenia brasiliensis Lam) é uma espécie de cereja nativa do Brasil que assim como as amora-preta (AP; Rubus spp) e silvestre (AS; Morus nigra L.) contém alto teor de antocianinas e elagitaninos. Poucos estudos focaram na ação anticâncer destas berries no câncer de mama. Portanto, o objetivo do presente estudo foi avaliar os efeitos de extratos ricos em antocianinas ou elagitaninos de G, AP e AS no crescimento e na expressão de genes e miRNAs das linhagens de células humanas de câncer de mama MCF-7 (receptor de hormônio positiva) e MDA-MB 231 (receptor de hormônio negativa). Não se observou citotoxicidade após 72 e 96 horas de tratamento com os extratos (25-200µg/mL) ricos em antocianinas (ASANT, APANT e GANT) ou elagitaninos (APELA e GELA), em ambas as linhagens celulares. Após 72 horas de tratamento, GANT e APANT induziram parada de ciclo celular em G0/G1 (12,5 e 50µg/mL, p<0,05) em células MCF-7. Após 96h, ASANT, APANT e GANT induziram parada de ciclo celular em G0/G1 (12,5 e 50µg/mL; p<0,05) nessas mesmas células. Entretanto, na concentração de 200µg/mL apenas GANT induziu parada em G0/G1 (72 e 96h; p<0,05). Em células MDA-MB 231, após 96h APANT e GANT induziram parada de ciclo celular em G0/G1 nas concentrações testadas (12,5, 50 e 200µg/mL, p<0,05), assim como ASANT nas concentrações de 12,5 e 50µg/mL (p<0,05). Em células MCF-7, após 72h APELA e GELA induziram aumento da proporção de células em subG0 (200µg/mL, p<0,05). Em MDA-MB 231, após 72 e 96h, APELA e GELA (200µg/mL) induziram aumento da proporção de células em subG0 (p<0,05) e parada em G0/G1 (p<0,05). Em células MCF-7, GANT induziu morte celular por apoptose (p<0,05) após 72 e 96h de tratamento. Entretanto, em MDA-MB 231 os extratos ricos em antocianinas não induziram morte celular. Em células MCF-7, após 96h GELA e APELA induziram principalmente necrose (p<0,05). Em MDA-MB 231, APELA e GELA induziram apoptose (p<0,05) após 72 e 96h. Em células MDA-MB 231, após 72h de tratamento foi observada inibição da proliferação celular por GELA, GANT e APELA (200µg/mL; p<0,05). Em células MDA-MB 231, 48h de tratamento com GELA; GANT e APELA (200µg/mL) aumentaram a expressão 5 genes (ESR2, FOXA1, JUN, PTGS2,VEGFA) e inibiram a expressão de 10 genes (ADAM23, ATM, BCL2, CDH1, EGF, GLI1, ID1, MKI67, SNAI2 e THBS1) correlacionados ao câncer de mama. Adicionalmente, GELA; GANT e APELA (200µg/mL) induziram aumento da expressão de miR- 210(p<0,05) e APELA (200µg/mL) reduziu a expressão de miRNA 19a/b (p<0,05) em células MDAMB 231. Coletivamente estes resultados sugerem que antocianinas de grumixama e elagitaninos de amora preta e grumixama apresentam potencial efeito protetor contra o câncer de mama. Adicionalmente, essa ação anticarcinogênica pode ser mediada por indução de morte celular, mais especificamente apoptose, redução de proliferação celular e modulação da expressão de genes e miRNAs relacionados ao câncer de mama.


Breast cancer is characterized as the neoplasia with the highest incidence and mortality rates in women worldwide. Anthocyanins and ellagitannins present in certain fruits, such as berries, stand out for their promising protective effect at different stages of breast cancer development. Grumixama (G; Eugenia brasiliensis Lam), a cherry species from Brazil, as well as blackberry (AP; Rubus spp) and mulberry (AS; Morus nigra L.) contain elevated concentrations of anthocyanins and ellagitannins. Few studies focused on the anticarcinogenic action of these berries in breast cancer development. Therefore, the aim of the present study was to evaluate the effects of extracts rich in anthocyanins or ellagitannins from G, AP and AS on cellular growth and genes and miRNAs expression in human MCF-7 (hormone receptor positive) and MDA-MB 231 (hormone receptor negative) cell lines. No cytotoxicity was observed after 72 and 96 hours of treatment with extracts rich in anthocyanin (25-200 µg/mL) (ASANT, APANT and GANT) or ellagitannins (APELA and GELA) in both cell lines. After 72 hours of treatment, GANT and APANT induced cell cycle arrest at G0/G1 (12.5µg/mL and 50µg/mL, p<0.05) in MCF-7 cells. After 96h, ASANT, APANT and GANT induced cycle arrest at G0/G1 (12.5 and 50µg/mL; p<0.05) in MCF-7 cells. However, at 200µg/mL, only GANT induced G0/G1 (72 and 96h; p<0.05). In MDA-MB 231 cells, after 96h APANT and GANT induced cell cycle arrest at G0/G1 with the three tested concentrations (12.5, 50 and 200µg/mL, p<0.05), as well as ASANT at concentrations 12,5 and 50µg/mL (p <0.05). In MCF-7 cells, after 72h APELA and GELA induced an increase in the proportion of cells in subG0 (200µg/mL, p<0.05). In MDA-MB 231, after 72 and 96h, APELA and GELA (200µg/mL) induced an increase in the proportion of cells in subG0 (p<0.05) and cell cycle arrest in G0/G1 (p<0.05). In MCF-7 cells, GANT induced apoptosis (p<0.05) after 72 and 96h of treatment. However, in MDA-MB 231, extracts rich in anthocyanins did not induce cell death. In MCF-7 cells, after 96h GELA and APELA induced mainly necrosis (p<0.05). In MDA-MB 231, APELA and GELA induced apoptosis (p<0.05) after 72 and 96h. In MDA-MB 231 cells, inhibition of cell proliferation by GELA, GANT and APELA (200?g/mL; p<0.05) was observed after 72h of treatment. In MDA-MB 231 cells, treatment for 48h with GELA, GANT and APELA (200µg) increased expression of 5 genes (ESR2, FOXA1, JUN, PTGS2, VEGFA) and inhibited expression of 10 genes (ADAM23, ATM, BCL2, CDH1, EGF, GLI1, ID1, MKI67, SNAI2 and THBS1) correlated with breast cancer. In addition, GELA; GANT and APELA (200µg/mL) induced increased expression of miR-210 (p<0.05) and APELA (200µg/mL) reduced the expression of miRNA 19a/b (p<0.05) in MDA-MB cells 231. Collectively these results suggest that anthocyanins of grumixama and ellagitannins of blackberry and grumixama have potential protective effect against breast cancer. Additionally, this anticarcinogenic action can be mediated by induction of cell death, more specifically apoptosis, reduction of cell proliferation and modulation of the expression of genes and miRNAs related to breast cancer.

ABSTRACT: Blackberry vinegar was produced in successive acetification cycles and content of total phenolics, anthocyanins and antioxidant activity were evaluated along the production. Firstly, blackberry wine was obtained in bench-scale bioreactor, being verified 0.39 g/g ethanol yield, 1.78 g/L.h volumetric productivity and 76% efficiency. After, three successive acetification cycles were conducted efficiently in grapia barrel with average acetic acid production of 51.6 g/L, 72.2 % acetic acid yield and 0.4 g/L.h volumetric productivity. Appreciable contents of polyphenolic compounds, anthocyanins and high antioxidant activity were observed in the raw material, wine and vinegar obtained in each cycle of acetic acid transformation. Acetic acid transformation led the small reduction of antioxidant activity compared to alcoholic fermentation, but the antioxidant potential was maintained along the cycles. The content of total phenolics and anthocyanins also suffered a reduction in step of acetification.

The present study aimed to study the preliminary phytochemical analysis, standardisation and in vitro antioxidant evaluation of Hippophae salicifolia (HS) berries extracts. Briefly, the berries were subjected to alcoholic and aqueous extraction. The preliminary phytochemical analysis was performed for aqueous and alcoholic extracts. The aqueous and alcoholic extracts are standardised for total phenolic, total tannin and total flavonoid content. The extracts were evaluated for its in-vitro free radical scavenging activities against, 2’-diphenyl-1- picrylhydrazyl (DPPH) radicals, hydroxyl radicals, nitric oxide radicals and total antioxidant activity. We found that alcoholic extract showed the presence of alkaloids, phenols,tannins, saponins, flavonoids and aqueous extract showed the presence of flavonoids, tannins, carbohydrates, and phenols. The extracts of HS berries were standardized for their total phenolic, tannin, flavonoid and vitamin-C content. The in vitro antioxidant activity revealed that both the extracts showed significant scavenging activities against free radicals. It can be concluded that both the extracts are effective in scavenging the free radicals. The flavonoids, tannins, vitamin-c and saponins may be attributed to free radical quenching activity of Hippophae salicifolia.

Las bayas de goji son frutas de un color rojo intenso, aproximadamente del tamaño de una pasa de uva y con un sabor que podría asemejarse a una mezcla de arándanos y cerezas. Las bayas frescas tienen un aspecto similar,tanto en la forma como en el tamaño, a la de los tomates cherry. China es el principal proveedor de bayas de gojiy se cultiva mayoritariamente en la provincia de Ni ngxia. Se comercializan principalmente desecadas, pero en elmercado también se encuentran jugos, aceites, extractos, compotas, mermeladas o jaleas, barras de cereal,muesli, y una gran variedad de productos cosméticos. Se les atribuyen numerosas propiedades nutricionales, pero debe destacarse que muy pocas han sido comprobadas científicamente. Objetivos del trabajo: Estudiar las características nutri cionales y organolépticas de las bayas de goji y de productos alimenticios elabo-rados con ellas, compararlos con los mismos productos sin bayas. Cuantificar los macronutrientes y antioxidan-tes, realizar la valoración nutricional, y determinar la aceptabilidad y satisfacción de los alimentos elaborados. Metodología: Este estudio es descriptivo y asimismo, corresponde a un diseño mixto, ya que p resenta una etapa no experi-mental-transeccional-descriptiva y en otra etapa, el diseño es cuasi-experimental. Resultados y conclusiones: Los alimentos que se elaboraron exitosamente con y sin bayas de goji fueron barritas de cereal y yogur helado, que resultaron con características organolépticas satisfactorias. Los productos elaborados con bayas fueron losque mayor contenido de carbohidratos y prote ínas contenían. En el caso de los lípidos, no se observaron grandes diferencias entre los productos elaborados con y sin bayas. Todos los alimentos presentaron fibra alimentaria en su composición. Goji berries are intense red-colored fruitsand approximately the size of a raisin. Its taste could be described as a combination of blueberries and cherries.Fresh goji berries are similar in form as well as in size tocherry tomatoes. China is the main supplier of goji berries, which are mostly harvested in the province of Ningxia. They are primarily marketed as dried fruits, but you can also find juices, oils, extracts, compotes, jams or jellies, cereal bars, muesli and a great variety of cosmetic products. Many nutritional properties are attributed to them, but it is worth noting that only a few of them have been proven scientifically. Objectives: To study the nutritional and sensory propertiesof goji berries and of food products made with them, and to compare these products with those that don't contain goji berries. To quantify the macronutrients and antioxidants, evaluate the nutritional value and determine theacceptability and satisfaction of the finished products. Methodology: This is a descriptive study with a mixed design, since it combines a non-experimental, descriptive and transactional phase with a quasi experimental phase. Results and conclusions: The foods that were successfullymade with and without goji berries were cereal bars and frozen yogurt, resulting in satisfactory sensory properties.The products made with goji berries had a higheramount of carbohydrates and proteins. As regards lipids, there were no significant differences between the products made with and without goji berries. All products had dietary fiber in their composition. As bagas de goji são frutas de cor vermelha intensa, aproximadamente do tamanho de uma uva passa e com um sabor que poderia assemelhar-se ao de uma mistura de blue berry e cerejas. As bagas frescas têm um aspecto similar, tanto na forma como no tamanho, às do tomate cereja. A China é o principal fornecedor de bagas de goji e são cultivadas em sua maioria na p rovíncia de Ningxia. São comercializadas principalmente dessecadas, mas no mercado também se encontram sucos, óleos, extratos, compotas, marmeladas, geleias barra de cereal, musli e uma grande variedade de produtos cosméticos. São atribuídas a elas numerosas propriedades nutricionais, mas é preciso ressaltar que muito poucas foram comprovadas cientificamente. Objetivos do trabalho: Estudar as características nutricionais e organolépticas das bagas de goji e de produtos alimentícios elaborados com elas compará-los com os mesmos produtos sem bagas. Quantificar os macronutrientes e antioxidantes, realizar a valorização nutricional, e determinar a aceitabilidade e satisfação dos alimentos elaborados. Metodologia: Este estudo é descritivo e também, corresponde a um desenho misto, já que apresenta uma etapa não experimental-transversal-descritiva e em outra etapa, o desenho é quase experimental. Resultados e conclusões:Os alimentos que foramelaborados com sucesso com e sem bagas de gofi foramas barrinhas de cereais e iogurte gelado, que resultaram com característica organolépticas satisfatórias. Os produtos elaborados com bagas foram os que apresentaram maior conteúdo de carboidrato e proteínas. No caso dos lipídios, não foram observadas grandes diferenças entre os produtos elaborados com e sem bagas. Todos os alimentos apresentaram fibra alimentar em sua composição. ABSTRACT

Objective: To investigate the chemical constituents from the berries of Physalis pubescens. Methods: The chemical constituents were isolated by repeated silica gel column chromatography, Sephadex LH-20 gel columns chromatography, medium pressure column chromatography, and semi-preparative liquid chromatography, and their structures were elucidated by chemical properties and spectral analyses. Results: Sixteen compounds were isolated and identified to be 3, 7, 3'-trimethylquercetin (1), kaempferol (2), chrysoeriol (3), quercetin-3-O-β-D-glucopyranoside (4), arjunolic acid (5), pulsatilla saponin A (6), pulsatilla saponin D (7), caffeic acid (8), 1-O-caffeoyl-β-D-glucopyranoside (9), N-trans-ferulogltyramine (10), neo-olivil (11), medioresinol (12), pinoresinol (13), sucrose (14), uridine (15), and β-sitosterol (16). Conclusion: Compounds 5-7 and 9-15 are obtained from the plants of genus Physalis L. for the first time. Compounds 3 and 8 are obtained from this plant for the first time.

Aims: To investigate if the immunomodulating activity of compounds present in berries and flowers of S. nigra were of the same order, or different, and also if the most active components were of high or low molecular weight nature. Methodology: Defatted material of berries and flowers of S. nigra were extracted with 50% ethanol and with water of 50ºC and 100ºC. High molecular weight fractions were obtained after gel filtration on BioGelP6DG. The different fractions were investigated for their monosaccharide contents and carbohydrate structures. The immunomodulating effects were investigated using a complement fixing assay as well as a system for measuring the production of NO after stimulation of macrophages with the different fractions. Results: All fractions contained substantial amounts of carbohydrates. Removal of low molecular weight material revealed polysaccharide fractions containing monosaccharides typical for pectins and showed enhanced bioactivity. Conclusion: High molecular weight fractions from elderflowers showed higher bioactivity than the equally extracted fractions from elderberries. The 100ºC water flower fraction gave after gel filtration the fraction with the highest activity and with the longest backbone of rhamnogalacturonan I. Based on this; one should encourage the drinking of juice made form flowers in the same way as has been done for the juice made from berries, since both may have a beneficial heath effect.

We hypothesize that the Ferric Reducing Antioxidant Power (FRAP) assay that follows the reaction of Fe3+-TPTZ at 593 nm underestimates the antioxidant capacity of fruits, since the standardized time of the reaction (4 min) is not enough to titrate all the reducing compounds available. We measured FRAP, total phenolics and anthocyanins content in a variety of Chilean berry fruits (blueberries, blackberries, raspberries and strawberries) and apples (cv. Fuji, Granny Smith, Pink Lady, Red Delicious and Royal Gala). Taking into account the dependence of FRAP on the time course of the reaction, we propose to measure FRAP indexes after 1 min (FRAP-1), 30 min (FRAP-30) and 120 min (FRAP-120) of incubation. Most fruit extracts showed significant correlations between the antioxidant capacity and the incubation time, although in some cases the FRAP indexes did not correlate with the total phenolics and/or anthocyanins content. In fact, in apples and berries the correlation between anthocyanins content and FRAP indexes decreased with the incubation time. It is concluded that the fruit extracts analyzed require an incubation period higher than the established in the original experimental protocol to reach the equilibrium, due to the presence of a complex mixture of antioxidant compounds. In addition, a kinetic profile should be realized in each sample studied to establish the most suitable incubation period to titrate all the reactive antioxidant species.


Se plantea que el ensayo de la capacidad antioxidante de frutas, medido según el poder reductor de hierro (FRAP), que sigue la reacción de Fe3+-TPTZ a 593 nm, subestima la capacidad antioxidante, debido a que el tiempo de reacción (4 min) no sería suficiente para que reaccionen todos los compuestos reductores disponibles en las muestras. Se analizó la capacidad antioxidante FRAP, el contenido de fenoles y de antocianinas en diversos berries (arándano, mora, frambuesa y frutilla) y manzanas (cv. Fuji, Granny Smith, Pink Lady, Red Delicious y Royal Gala). Tomando en cuenta la dependencia del tiempo de incubación en el valor FRAP, se propone medir los índices FRAP después de 1 min (FRAP-1), 30 min (FRAP-30) y 120 min (FRAP-120). Diversos extractos de las frutas analizadas mostraron una correlación significativa entre la capacidad antioxidante y el tiempo de incubación; sin embargo, en algunos casos los índices FRAP no se correlacionaron con el contenido de fenoles totales y/o antocianinas. En efecto, en manzanas y berries la correlación entre el contenido de antocianinas e índices FRAP disminuyó con el tiempo de incubación. Se concluye que los extractos analizados requieren un tiempo de incubación mayor al que establece el protocolo analítico original para alcanzar el equilibrio, debido a la presencia de una compleja mezcla de compuestos antioxidantes. Además, el perfil cinético de cada muestra debería ser estudiado para establecer el periodo de incubación más adecuado para titular todas las especies antioxidantes reactivas.
